Biological Activity of ON-01910:
ON 01910.Na is a Polo-like kinase 1 inhibitor. It is a synthetic benzyl styryl sulfone analogue with potential antineoplastic activity. It inhibits Plk1 pathway activity at 9–10 nM in a substrate-dependent andATP-independent manner. It induces selective G2/M arrest followed by apoptosis in a variety of tumor cells while causing reversible cell arrest at the G1 and G2 stage without apoptosis in normal cells. This agent may exhibit synergistic antitumor activity in combination with other chemotherapeutic agents. In vitro,ON01910.Na exhibited a broad spectrumof activity, with IC50 values in the range of 50–250 nM in100 different cell lines tested. [1,2,3]
